چکیده

Phenoloxidases (EC.1.14.18.1) are implicated in the immunity of insects toward microorganisms, so oxidative enzymes such as phenoloxidases (PO) from 7th instar larvae red palm weevil; Rhynchophorus ferrugineus (Oliv.) was partially isolated to characterize its activity using catechol a substrate. The mitochondrial fraction larval homogenate used enzyme source for partial kinetic studies.Enzyme catalysis directly proportional crude protein conc. up 200 ug contained 1 ml reaction mixture. Michaelis-Menten kinetics PO evaluated by constructing Lineweaver Burk double reciprocal plot. Michaelis constant (Km) was, 3.45 X10-4 M and maximum velocity (V max) 344.8 O.D.min-1 mg protein-1. Effect pH, temperature substrate concentration on tested. Generally, each variable chosen while other conditions were at optimum found primarily. Reaction rate optimal 10-1 N catechol, neutral medium (pH 7) 40 C. kept most when incubated 60 C 15 min before initiation. efficiently catalyze catechol. Results present study may probably offer fundamental anchor future alternative strategies controlling R. via obstructing innate

Digestive proteases during development of larvae of red palm weevil, Rhynchophorus ferrugineus (Olivier, 1790) (Coleoptera: Curculionidae).

The evolution of digestive proteases during larval development of Rhynchophorus ferrugineus (Olivier, 1790) has been studied. A progressive increase of protease activity has been found. The optimum pH for proteolytic activity against azocasein was determined. Caseinograms revealed an active complex of alkaline proteases from the early stages of the development. From the apparent molecular masse...

Screening of tropical isolates of Metarhizium anisopliae for virulence to the red palm weevil Rhynchophorus ferrugineus Olivier (Coleoptera: Curculionidae)

The red palm weevil, Rhynchophorus ferrugineus Olivier (Coleoptera: Curculionidae) is a serious pest of the palm tree in tropical regions of the world. One strain of Metarhizium sp. ZJ-1, isolated from Chinese soils, was evaluated for growth characteristics, and screened for its virulence to R. ferrugineus larvae in laboratory conditions. An approximately 685-bp fragment was amplified by ITS (I...

The Gut Entomotype of Red Palm Weevil Rhynchophorus ferrugineus Olivier (Coleoptera: Dryophthoridae) and Their Effect on Host Nutrition Metabolism

For invasive insects, the potential roles of gut microbiota in exploiting new food resources and spreading remain elusive. Red palm weevil (RPW), Rhynchophorus ferrugineus Olivier, is an invasive destructive pest which feeds on nutrient-poor tender tissues and has caused extensive mortality of palm trees. The microbes associated with insects can improve their nutrition assimilation. However, ex...

A meridic diet for laboratory rearing of Red Palm Weevil, Rhynchophorus ferrugineus (Coleoptera: Curculionidae)

The red palm weevil (RPW) Rhynchophorus ferruginous (Olivier) was fully developed and reared on a meridic diet consisting of agar, distilled water, commercial yeast as well as laboratory produced amino and fatty acid rich brewer’s yeast (Saccharomyces cerevisiae), wheat meal, corn flour, benzoic acid, ascorbic acid, sorbic acid, vitamin mix and tetracycline hydrochloride. A group of weevils rea...

Identification of pigmented Serratia marcescens symbiotically associated with Rhynchophorus ferrugineus Olivier (Coleoptera: Curculionidae)

To characterize red pigment-producing bacteria (RPPB) regularly released during oviposition by red palm weevil (RPW), RPPB were recovered from eggs deposited in apples supplied as substrate for oviposition. The presence of RPPB was also detected from gut, the reproductive apparatus of dissected adult and virgin insects and from pupal cases collected within infested palms. RPPB were also identif...
